    An alternating electric field between spatially different pairs of electrodes causes megabase (Mb) DNA to reorient and move at different speeds through pores in an agarose gel. The voltage changes periodically between three directions; one passing through the central axis of the gel and two extending at a 60-degree angle on either side. Pulse times are the same for each direction resulting in net DNA migration forward. For extremely large molecules, switching interval ramps can be used that increase the pulse time for each direction over a specified number of hours. The CHEF Mapper system separates large and small DNA fragments with better resolution, speed and accuracy than traditional field pulsation methods. The method effectively separates DNA sizes ranging from 100 pb to over 10 Mb.
